Verbinski believes the constant shooting aided in the intimate feeling on set. This caused much of the backgrounds and certain ships to be fully created in post-production using CGI. To keep on schedule, Verbinski and crew had to constantly be shooting. They were all influenced and mentored by Hans Zimmer and Klaus Badelt who came up with the basic themes used throughout. Verbinski explains how 15 different composers worked on the music for Curse of the Black Pearl.Depp recalls wanting to include a line of dialogue where Sparrow speculates Will Turner to be a eunuch. Verbinski recalls how at certain times, if there was something mentioned the screenwriters didn’t like, the actors would go ahead and do it anyway. This allowed for the cast to constantly throw out ideas and improvisations for their characters that could end up being written into the script. Screenwriters Ted Elliott and Terry Rossio were on set every day of filming, a rarity for film shoots, especially those this size and a first for these particular screenwriters.“You’re still a dork,” Verbinski would say to him. ![]() He wanted to play the character more like Depp’s Jack Sparrow, but Verbinski had to keep reiterating to him how uncool Will Turner is. This DVD offers three separate commentaries featuring various members of the cast and crew, but rather than hear the insight Jack Davenport had to offer – we love you, Jack – it’s probably best to hear from the film’s director and star. We’re not scraping the bottom of the barrel just yet, and even though Curse of the Black Pearl is by no means a bad movie, it just hasn’t reached a level of beloved nostalgia like our first two. ![]() Especially when you consider this week’s Commentary Commentary, our third, goes from essential classics like The Thing and Die Hard to Pirates of the Caribbean: The Curse of the Black Pearl. The outer Grenadine island of Petite Tabac became the deserted isle where Jack Sparrow ( Johnny Depp) and Elizabeth Swann ( Keira Knightley) are abandoned by Captain Barbossa ( Geoffrey Rush). The harbour of ‘Port Royal’ was built at Wallilabou Bay on St Vincent (the dock and a facade have been left), and the small town set (which has now gone) at Chateaubelair. St Vincent in the Grenadines, part of the Windward Islands in the south Caribbean, was the production base in the southern Caribbean. The Governor’s mansion was also built here (the interior being a set at Raleigh Studios in Manhattan Beach, a little further north up the coast). ‘Fort Charles, Port Royal’ was built on a three-acre bluff on the Palos Verdes Peninsula, south of Los Angeles, on the site of Marineland, a one-time amusement park (near the Point Vicente Lighthouse seen in Pearl Harbor). Yeah, I am not blind to the irony of writing a book based on healthy food choices while my own waistline expanded a bit beyond my ideal weight range. For example, the genome-wide association studies (GWAS) have identified genetic loci associated with longevity and several aging-related diseases. Over the last decade, there has been a growing body of research in identifying genetic and epigenetic biomarkers of aging to decipher the molecular mechanisms underpinning disease susceptibility. Increasing evidence has pointed to the interactions between genetics, epigenetics and environmental factors in the aging process. RNAAgeCalc is available at, both as Bioconductor and Python packages, accompanied by a user-friendly interactive Shiny app.Īging is among the most complex phenotype and is a well-known risk factor for a myriad of diseases including cardiovascular, diabetes, arthritis, neurodegeneration and cancer. Furthermore, we demonstrate that the transcriptional age acceleration computed from our within-tissue predictor is significantly correlated with mutation burden, mortality risk and cancer stage in several types of cancer from the TCGA database, and offers complementary information to DNA methylation age. Our results also indicate that both racial and tissue differences are associated with transcriptional age. We show that our transcriptional age calculator outperforms other prior age related gene signatures as indicated by the higher correlation with chronological age as well as lower median and median error. Based on these genes, we develop new across-tissue and tissue-specific age predictors. By performing a meta-analysis of transcriptional age signature across multi-tissues using the GTEx database, we identify 1,616 common age-related genes, as well as tissue-specific age-related genes. Here, we introduce RNAAgeCalc, a versatile across-tissue and tissue-specific transcriptional age calculator. ![]() Numerous epigenetic age calculators are available, however biological aging calculators based on transcription remain scarce. Biological aging reflects decline in physiological functions and is an effective indicator of morbidity and mortality. ![]() |
